Human Gene SPHK1 (ENST00000592299.5) Description and Page Index
Description: Homo sapiens sphingosine kinase 1 (SPHK1), transcript variant 3, mRNA. (from RefSeq NM_001142601) RefSeq Summary (NM_001142601): The protein encoded by this gene catalyzes the phosphorylation of sphingosine to form sphingosine-1-phosphate (S1P), a lipid mediator with both intra- and extracellular functions. Intracellularly, S1P regulates proliferation and survival, and extracellularly, it is a ligand for cell surface G protein-coupled receptors. This protein, and its product S1P, play a key role in TNF-alpha signaling and the NF-kappa-B activation pathway important in inflammatory, antiapoptotic, and immune processes. Phosphorylation of this protein alters its catalytic activity and promotes its translocation to the plasma membrane. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Sep 2017]. ID:SPHK1_HUMAN DESCRIPTION: RecName: Full=Sphingosine kinase 1; Short=SK 1; Short=SPK 1; EC=18.104.22.168; FUNCTION: Catalyzes the phosphorylation of sphingosine to form sphingosine 1-phosphate (SPP), a lipid mediator with both intra- and extracellular functions. Also acts on D-erythro-sphingosine and to a lesser extent sphinganine, but not other lipids, such as D,L-threo-dihydrosphingosine, N,N-dimethylsphingosine, diacylglycerol, ceramide, or phosphatidylinositol. CATALYTIC ACTIVITY: ATP + sphinganine = ADP + sphinganine 1- phosphate. CATALYTIC ACTIVITY: ATP + sphingosine = ADP + sphingosine 1- phosphate. COFACTOR: Magnesium. BIOPHYSICOCHEMICAL PROPERTIES: Kinetic parameters: KM=14 uM for sphingosine; KM=20 uM for dihydrosphingosine; KM=77 uM for ATP; pH dependence: Optimum pH is 7.4; SUBUNIT: Interacts with ACY1 (By similarity). Binds to calmodulin. Interacts with SPHKAP. INTERACTION: P68104:EEF1A1; NbExp=2; IntAct=EBI-985303, EBI-352162; Q14192:FHL2; NbExp=7; IntAct=EBI-985303, EBI-701903; Q2M3C7:SPHKAP; NbExp=4; IntAct=EBI-985303, EBI-1803914; SUBCELLULAR LOCATION: Cytoplasm. Nucleus. TISSUE SPECIFICITY: Widely expressed with highest levels in adult liver, kidney, heart and skeletal muscle. MISCELLANEOUS: Sphingosine 1-phosphate stimulates TRAF2 E3 ubiquitin ligase activity, and promotes activation of NF-kappa-B in response to TNF signaling. SIMILARITY: Contains 1 DAGKc domain.
The RNAfold program from the Vienna RNA Package is used to perform the secondary structure predictions and folding calculations. The estimated folding energy is in kcal/mol. The more negative the energy, the more secondary structure the RNA is likely to have.
ModBase Predicted Comparative 3D Structure on Q9NYA1
The pictures above may be empty if there is no ModBase structure for the protein. The ModBase structure frequently covers just a fragment of the protein. You may be asked to log onto ModBase the first time you click on the pictures. It is simplest after logging in to just click on the picture again to get to the specific info on that model.
Orthologous Genes in Other Species
Orthologies between human, mouse, and rat are computed by taking the best BLASTP hit, and filtering out non-syntenic hits. For more distant species reciprocal-best BLASTP hits are used. Note that the absence of an ortholog in the table below may reflect incomplete annotations in the other species rather than a true absence of the orthologous gene.
