Human Gene GNL3 (ENST00000418458.6) from GENCODE V44
  Description: Homo sapiens G protein nucleolar 3 (GNL3), transcript variant 1, mRNA. (from RefSeq NM_014366)
RefSeq Summary (NM_014366): The protein encoded by this gene may interact with p53 and may be involved in tumorigenesis. The encoded protein also appears to be important for stem cell proliferation. This protein is found in both the nucleus and nucleolus. Three transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Nov 2010].

DESCRIPTION: RecName: Full=Guanine nucleotide-binding protein-like 3; AltName: Full=E2-induced gene 3 protein; AltName: Full=Novel nucleolar protein 47; Short=NNP47; AltName: Full=Nucleolar GTP-binding protein 3; AltName: Full=Nucleostemin;
FUNCTION: May be required to maintain the proliferative capacity of stem cells. Stabilizes MDM2 by preventing its ubiquitination, and hence proteasomal degradation (By similarity).
SUBUNIT: Interacts with MDM2; this interaction stabilizes MDM2. Interaction with MDM2 occurs in the nucleoplasm and is triggered by a nucleolar release mechanism, such as mitosis-induced nucleolar disassembly (By similarity). Indirectly interacts with TP53, via MDM2-binding (By similarity).
INTERACTION: Q00987:MDM2; NbExp=3; IntAct=EBI-641642, EBI-389668; Q15172:PPP2R5A; NbExp=3; IntAct=EBI-641642, EBI-641666; P40763:STAT3; NbExp=2; IntAct=EBI-641642, EBI-518675;
SUBCELLULAR LOCATION: Nucleus (By similarity). Nucleus, nucleolus (By similarity). Note=Shuttles between the nucleus and nucleolus (By similarity).
TISSUE SPECIFICITY: Increased levels in lung tissue in cancer patients.
DOMAIN: The basic domain (B) allows nucleolar localization in the absence of GTP. The intermediate domain (I) inhibits nucleolar localization by the B domain and is required for exit from the nucleolus. Exit from the nucleolus to the nucleoplasm requires both the I and the acidic (A) domains, and may be triggered by GTP hydrolysis (By similarity).
DOMAIN: In contrast to other GTP-binding proteins, this family is characterized by a circular permutation of the GTPase motifs described by a G4-G1-G3 pattern.
SIMILARITY: Belongs to the MMR1/HSR1 GTP-binding protein family.
SIMILARITY: Contains 1 G (guanine nucleotide-binding) domain.

The RNAfold program from the Vienna RNA Package is used to perform the secondary structure predictions and folding calculations. The estimated folding energy is in kcal/mol. The more negative the energy, the more secondary structure the RNA is likely to have.

The pictures above may be empty if there is no ModBase structure for the protein. The ModBase structure frequently covers just a fragment of the protein. You may be asked to log onto ModBase the first time you click on the pictures. It is simplest after logging in to just click on the picture again to get to the specific info on that model.

-  Orthologous Genes in Other Species
  Orthologies between human, mouse, and rat are computed by taking the best BLASTP hit, and filtering out non-syntenic hits. For more distant species reciprocal-best BLASTP hits are used. Note that the absence of an ortholog in the table below may reflect incomplete annotations in the other species rather than a true absence of the orthologous gene.
